As has already been stated in response to this same question in another thread, swapping to 150% larger injectors is considered a "major" upgrade.
Look at a "minor" upgrade, such as an intake. The engine will run fine (not necessarily great) without tuning to compensate.
Try dropping in the green giants without tuning and you will discover just how "major" it really is.

It was the 42# injector pn thread, I believe. I would post a link, but its a pita from the phone, and it wouldn't answer your question anyway.
Regarding the tune update, I think you have two options: 1) order it through whichever avenue you purchased the premium tune in the first place, or 2) contact Vince at trifecta directly.
I have never updated my tune, so im not speaking from experience, just relaying what I've gleaned from the forum over the past year+. Hell, I could be completely off.

Submit a ticket at BNR for the upgraded injector tune or pm him, then he will get you straight.

not sure why he is but if you are running wot for long periods of time.. Like doing track. the stock ones run max duty cycle and you could start missing. I have done this on my track sessions I didn't realize what it was at first.. I will be instaliing them as soon as i get my water/meth straight..
Injectors are going static at current power level on pump gas(188whp/237wtq ). Injector pulsewidth peaks at 27ms and never drops below 20ms so they are at 100% from about 4,000 until 6,500. Obviously this isn't safe, so big injectors are going in, plus it does give me and E-85 option.
I hear you at that power level you need them.With my tune i am at 178HP/201TQ flywheel i think my injectors are around 85% duty cycle at 6000 rpm.So in my case i would not need them.If i did anything else to mine it would just be meth injection.I do think your car would really benefit from meth.
